The metal-responsive element (MRE)-binding transcription factor (MTF-1) stimulates the expression of metallothioneins in response to the exposure of cells to heavy metals. MTF-1 contains six zinc fingers in the DNA binding domain. The phosphorylation of MTF-1 in response to metal exposure appears to play a significant role in the ability of MTF-1 to activate metallothionein transcription. In addition to its role in metallothionein activation, MTF-1 is involved in a post-transcription regulatory complex for ribosomal protein S25. MTF-1, La and p53 inhibit the nuclear export of S25 mRNA in response to nutrient depravation. Furthermore, MTF-1 acts as a chromatin insulator on integrated transgenes in cultured cells to insulate active loci against chromatin silencing. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms. Within the desmosome, the extracellular domain of desmoglein is essential for calcium dependent heterophilic binding to the desmocollins, whereas the intracellular domain is essential for binding to the desmosomal plaque protein, plakoglobin. This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Galectins are a family of soluble beta-galactoside-binding lectins that modulate cell-to-cell adhesion and cell-to-extracellular matrix (ECM) interactions and play a role in tumor progression, pre-mRNA splicing and apoptosis. One such member, galectin-13, is a 139 amino acid protein that contains one galectin domain. Possessing lysophospholipase activity, galectin-13 exists as a disulfide-linked homodimer. Galectin-13 is suggested to have a developmental role in the placenta and may display immunobiological activity in feto-maternal blood-spaces.
